Selective strand scission by intercalating drugs at DNA bulges.
A bulge is an extra, unpaired nucleotide on one strand of a DNA double helix. This paper describes bulge-specific strand scission by the DNA intercalating/cleaving drugs neocarzinostatin chromophore (NCS-C), bleomycin (BLM), and methidiumpropyl-EDTA (MPE). For this study we have constructed a series of 5'-32P end labeled oligonucleotide duplexes that are identical except for the location of a b...
متن کاملInterference of intrinsic curvature of DNA by DNA-intercalating agents.
It has been demonstrated in our studies that the intrinsic curvature of DNA can be easily interrupted by low concentrations of chloroquine and ethidium bromide. In addition, the changes of DNA curvature caused by varying the concentration of these two DNA intercalators can be readily verified through using an atomic force microscope.
متن کاملAttenuation of Cytotoxic Natural Product DNA Intercalating Agents by Caffeine
Many anti-tumor drugs function by intercalating into DNA. The xanthine alkaloid caffeine can also intercalate into DNA as well as form π-π molecular complexes with other planar alkaloids and anti-tumor drugs. The presence of caffeine could interfere with the intercalating anti-tumor drug by forming π-π molecular complexes with the drug, thereby blocking the planar aromatic drugs from intercalat...
متن کاملActivation of cGAS-dependent antiviral responses by DNA intercalating agents
Acridine dyes, including proflavine and acriflavine, were commonly used as antiseptics before the advent of penicillins in the mid-1940s. While their mode of action on pathogens was originally attributed to their DNA intercalating activity, work in the early 1970s suggested involvement of the host immune responses, characterized by induction of interferon (IFN)-like activities through an unknow...
متن کاملDNA Intercalators and Using Them as Anticancer Drugs
Many anticancer drugs in clinical use interact with DNA through intercalation, which is process that starts with the transfer of the intercalating molecule from an aqueous environment to the hydrophobic space between two adjacent DNA base pairs. In general, intercalatig agents are two types: monofunctional and bifunctional. Monofunctional intercalators contain one intercalating unit and Bifunct...
